What a lot number is actually for
A lot number looks like an inventory detail. It is closer to the opposite: it is the only mechanism by which anything measured about a material can be attached to the material itself.
Without it, a purity figure is a claim about a product family. With it, a purity figure is a claim about a specific quantity of powder produced on a specific run, and that claim can be checked.
What a lot actually is
A lot is a quantity of material produced under one set of conditions in one continuous operation. For a synthetic peptide that generally means one synthesis, one purification campaign, and one lyophilisation run, filled and sealed together.
The boundary matters because it defines what a measurement covers. Testing a sample from a lot supports conclusions about that lot. It supports nothing about the next synthesis, even of the same sequence by the same route, because the variables that produce impurity profiles are run-specific: resin batch, coupling efficiency at each residue, cleavage conditions, how the gradient was run, where fractions were cut.
Two lots of the same compound at the same nominal purity can carry different impurity profiles. The headline number matches; what sits underneath it does not.
Why catalogue averages fail
When a supplier reports a single figure across a catalogue entry, that figure is one of two things: the result from one favourable lot, generalised, or an average across lots.
Both destroy the information that makes the number useful. An average tells you about a distribution while you hold one sample from it. A generalised best result tells you what was achievable once.
It also makes error invisible. If a lot goes wrong, per-lot reporting isolates it to that lot. Catalogue-level reporting smears it across everything sharing the entry, and there is no way afterwards to determine which vials were affected.
What traceability requires
For a lot number to mean anything, four things have to hold:
- The number is on the vial. Not only on a box or a packing slip, both of which get separated from the contents.
- The number is on the certificate, and the certificate reports figures measured on that lot rather than inherited from elsewhere.
- The number resolves in an archive. Quoting it to the supplier returns the underlying record: synthesis conditions, raw chromatograms, mass spectrometry data, the third-party certificate, the cold-chain log.
- The archive is retained. A record that is discarded after ninety days cannot answer a question raised in month five.
Any one of these missing breaks the chain. A number etched on a vial that resolves to nothing is decoration.
Where it earns its keep
Reproducibility
When work is repeated and something differs, material is one of the variables. Being able to say that both runs used the same lot removes it from consideration. Being able to say they used different lots, and to pull both impurity profiles, turns a mystery into something examinable.
Methods sections
Published work that cites a supplier and a catalogue number has specified a product family. Citing the lot number specifies the material. The difference is whether a reader can establish what was actually used.
Reordering
Where consistency matters across a longer piece of work, ordering from the same lot family, or checking a new lot's profile against the previous one before committing to it, is the mechanism. That is only possible if lots are tracked as distinct entities rather than collapsed into a catalogue entry.
Handling a problem
If something is wrong with a material, the first question is scope. Per-lot records answer it: which run, which quantity, which shipments. Without them the honest answer is that the boundary is unknown.
Questions worth asking a supplier
- Is the lot number etched or printed on the vial itself?
- Are the certificate figures measured on this lot, or inherited from the catalogue entry?
- Can raw chromatograms and mass spectrometry data be retrieved by lot number?
- How long are records retained after a lot is exhausted?
- Is third-party testing performed per lot, or periodically across lots?
- Is the cold-chain record for a shipment retrievable afterwards?
The answers separate suppliers who track material from suppliers who track products. Both can sell the same compound. Only one can tell you what is in the vial you are holding.
